Adequan for Dogs: Does It Work, and What the Label Actually Says

Adequan for dogs comes up most often as the other injection, the one a vet mentions after you have already read something frightening about Librela. It is older, it is given differently, and the way it is marketed and the way it is approved are not quite the same thing.

That last part is worth twenty minutes of your time before an appointment, because the gap is not a scandal and it is not nothing either. This walks through what the label says, what the trial behind it actually measured, who should be careful with it, and what the July 2026 recall does and does not mean for your dog.


The short version: Adequan Canine is an injectable prescription drug, approved by the FDA in 1997, given into the muscle twice a week for four weeks. Its approved use is the control of arthritis signs, and the dog trial supporting it found better range of motion and better overall orthopedic scores than saline. The widely repeated claim that it slows or reverses cartilage damage traces to laboratory work on cultured cells rather than to that trial. Two lines on the label matter more for an old dog than for a young one: it is a heparinoid, so it is not for dogs with bleeding disorders, and it comes with a caution for dogs with kidney or liver impairment. A four-lot recall in July 2026 covered specific lot numbers, not the product as a whole.


What Adequan actually is

Adequan Canine is polysulfated glycosaminoglycan, usually shortened to PSGAG. The package insert describes it as a semi-synthetic glycosaminoglycan prepared "by extracting glycosaminoglycans (GAGs) from bovine tracheal cartilage." It comes as a 100 mg/mL solution in a 5 mL multidose vial, and it goes into muscle rather than under the skin or into the joint.

It has been FDA-approved since 1997, which makes it one of the oldest arthritis drugs still in regular use in dogs, and it sits in a different category from the daily NSAIDs most senior dogs start on. Our guide to managing arthritis at home covers where each layer fits.

Does Adequan work for dogs?

Yes, in the specific sense that a blinded, placebo-controlled trial found a measurable benefit. It is worth knowing exactly how big that trial was and what it measured, because both are smaller than the reputation.

The insert describes the pivotal field trial in dogs with radiographically confirmed joint disease in one or two joints. "Fifty-one dogs were randomly assigned to receive either" the drug at 2 mg/lb of body weight or a 0.9% saline placebo. In the insert's words, "Both treatments were administered by intramuscular injection twice weekly for 4 weeks (8 injections total)." The vets giving the injections and scoring the dogs did not know which dogs got which. Each lame limb was scored for lameness at a walk, lameness at a trot, pain, range of motion, and functional disability, and those scores were combined into a total orthopedic score.

The result, in the label's own words, is that dogs treated with the drug showed a "statistically significant improvement in range of motion and total orthopedic score over placebo treated control dogs."

Three things about that are worth holding onto. Twenty-four dogs were in the treated group, which is a small trial by any standard. The dogs had one or two affected joints, not the whole-body arthritis many thirteen-year-olds are carrying. And what improved was how the dogs moved and how they scored on an orthopedic exam, which is a real and meaningful outcome and is also not the same as what the marketing says.

The cartilage claim, and where it comes from

The manufacturer's own homepage says Adequan Canine is "the only FDA-approved drug that has been shown to slow down and even reverse cartilage deterioration in a dog's joints." That sentence is why most owners have heard of the drug at all, and it is the reason it gets described as disease-modifying rather than pain-relieving.

The FDA-approved indication is narrower. The label says the drug is "recommended for intramuscular injection for the control of signs associated with non-infectious degenerative and/or traumatic arthritis of canine synovial joints." Control of signs. Not repair of cartilage.

So where does the cartilage language come from? From the pharmacology section of the same label, and that section is explicit about its setting. It reports that "Experiments conducted in vitro have shown PSGAG to inhibit certain catabolic enzymes which have increased activity in inflamed joints, and to enhance the activity of some anabolic enzymes." The cartilage-building side of that comes from cell culture too: "Cultured human and rabbit chondrocytes have shown increased synthesis of proteoglycan and hyaluronic acid in the presence of PSGAG."

Those passages describe real, published laboratory pharmacology, and the manufacturer is not inventing anything. What the label does not contain is a trial in living dogs that measured cartilage and found it preserved. The dog trial measured lameness, pain, and range of motion. The conclusion I draw from that pair of documents is my own rather than the label's: the mechanism is plausible, the clinical evidence in dogs is about comfort and movement, and the honest way to judge a course is whether your dog moves better on it rather than whether a joint is being rebuilt.

That distinction changes what a failed course means. If eight injections do nothing for how she gets up in the morning, there is no separate invisible benefit still accruing that justifies continuing.

Adequan canine dosage: what a course looks like

The manufacturer's dosing page gives the schedule in one line: "Administer twice weekly for up to 4 weeks (maximum of 8 injections) at the approved dosage." The dose itself is "2 mg/lb body weight (0.02 mL/lb or 1 mL/50 lbs) by intramuscular injection only."

Two practical consequences follow from that arithmetic, and they are the part nobody puts in a brochure.

Eight visits is a lot of visits. Twice a week for four weeks is a real burden for a dog who finds the car hard, which is why many vets teach owners to give the injections at home. If your dog is the kind who dreads the clinic, ask about that at the first appointment rather than after visit three.

The vial size decides your real cost. A 5 mL vial at 1 mL per 50 lb means one vial covers five injections for a 50 lb dog, or ten for a 25 lb dog, while a 90 lb dog uses close to two mL a time and needs the best part of three vials for a full course. When you ask a clinic what Adequan costs, ask for the price per vial and how many vials your dog's weight needs for a full course, because the per-injection number on its own tells you very little. Prices vary widely between clinics and pharmacies, so this is a question for yours rather than something to take from any website, this one included.

Adequan for dogs side effects

The trial's safety numbers are modest. The insert reports that in that study, "Possible adverse reactions were reported after 2.1% of the injections." What those were: transient pain at the injection site, transient diarrhea in two dogs, and one incident of abnormal bleeding. The label describes them as mild and self-limiting.

Two lines further up matter more for an older dog than for a young one, and they are easy to skim past.

It is a blood thinner's cousin. The label's contraindication reads: "PSGAG is a synthetic heparinoid; do not use in dogs with known or suspected bleeding disorders." That is a real consideration if your dog is on other medication, has a clotting problem, or has surgery coming up, including a dental. Raise it before the course starts rather than the week of.

Kidneys and liver. The precautions include a single sentence that lands differently in a geriatric patient: "Use with caution in dogs with renal or hepatic impairment." Kidney change is the kind of thing an older dog can be carrying quietly, which is the whole argument for senior bloodwork in the first place. If your dog has not had recent values run, that is the thing to line up alongside this conversation rather than after it.

The label also notes that safe use in breeding, pregnant, or lactating dogs has not been evaluated, which is rarely the situation in the dogs this site is about but belongs in an honest list.

Adequan vs Librela

This is the comparison most people arrive wanting, usually after reading about Librela's safety reports. The two are not competing versions of the same thing.

The practical differences follow from the schedule more than from the pharmacology. Adequan is a defined course with an end, which makes it easier to try and easier to abandon if nothing changes. Neither of them replaces the unglamorous layers: weight control, traction on your floors, and the supplements with actual evidence.

The 2026 Adequan recall, and whether it affects your dog

On July 31, 2026, American Regent announced a nationwide recall covering four lots, two of them Adequan Canine. The FDA published the notice on August 3. The reason, in the agency's posting of the company announcement, is that "The products are being recalled due to the presence of glass fibers found during testing of internally retained samples."

The canine lots named are 25011 and 3369, in the 100 mg/mL 5 mL multidose vial. The other two lots in the recall, 24416 and 25265P, are the equine version.

What this is: a defect in specific manufacturing lots. What it is not: a finding about whether the drug works or a new safety signal about PSGAG itself. If your dog is mid-course or you have a vial at home, the useful thing is to read the lot number off the vial or ask your clinic to check theirs, and to tell your vet if your dog had any reaction after an injection from one of those lots.

A dog in significant pain now is a different case from a dog with early changes, and the choice also depends on whether an ongoing commitment or a finite trial suits your situation.
